Police say the route reopened shortly after midnight on Saturday, about 19 hours after the crash.
The development could also contain land for a school, shops and a traveller site.
National Highways said the road would be closed in phases, between Gamston and Bingham, from Monday.
Local NHS bosses say the expansion is needed to support a growing population.
Police said there were not currently believed to be any serious injuries following the crash.
